Not sure I follow. Let's say the trim is not fully seated on the top of the glass on one side of the tank and you put a level across the trim (which is skewed in relation to the tank bottom). Now you put a level across the top of the stand (let's assume the trim on the bottom of the tank is on correctly). One of these two readings can show level, but not both. Either you're leveling according to the crooked trim or you're leveling according to the tank bottom/top of stand.
FWIW, I usually use a level of some kind to setup a tank, but I also check the water level all around periodically to make sure nothing has changed due to settling/shifting as the addition of the water increases the weight of the whole setup. Just not sure I'm following the idea mentioned above...
